Sensing the point of no return

When is it time to die, that is the question that lays behind the latest study of a group of Swedish researchers. They found that sensing depleted levels of the amino acid arginine tell the cell to throw in the towel.

Leaf senescence and subsequent dead are a normal part of a plant’s life. We see it in action every autumn when leaves turn red and yellow before they fall of the trees. It also happens during the normal development when older leaves die and during periods of stress like drought stress.

During leaf senescence cells degrade their proteins, nucleic acids, lipids, and cell structures to recycle the nutrients within. These they then transport out of the cell for use by younger organs or for storage. Although this doesn’t have to end in a cell’s death. If, like for example, in the case of drought stress the stress ends before the senescing cell has reached the point of no return the cell can recover.

Preventing to senescence

This begs the question: how does the cell know when it is time to die? Plant breeders see the answer to this question as a way to obtain more control over leaf senescence. This they hope will help reducing yield losses and allow the breeding of crops that are less sensitive to senescence inducing stress.

The first hint to the answer to this question the researchers got from a mutant that doesn’t initiate stress-induced senescence. The leaves of those plants accumulate large quantities of amino acids. To find out of any of these amino acids influence the senescence process the researchers exposed leaf discs to an extra amino acid. This showed that only in the case of extra arginine the leaf disc maintained more of its chlorophyll compared to discs treated with the other amino acids or the control ones. Suggesting that a reduction of arginine below threshold levels signals that it is time to senescence.

Restoring the ability to senescence

Next the researchers checked on genes that transport arginine out of the cytosol. Of two genes who did those jobs, AAP5 and BAC2, the researchers found that they were activated during senescence. Of these BAC2 was even activated directly by the senescence master regulator. Subsequently the researchers activated these arginine transporters in plants whose leaves don’t initiate senescence. Finding that this restored the ability to senescence.  

Now the important question was, does this also work the other way around. That is if when arginine levels stay high the cell delays senescence during normal aging. So, they created plants without AAP5 or BAC2 or both. While plants who missed only one of these arginine transporters did not delay their developmental aging induced senescence, those who missed both did.

So, the levels cytosolic arginine likely functions as a way to tell the cell at what stage of senescence it is at. But like the researchers are saying more details are needed. Like if this does depend how cytosolic arginine is distributed. Or like how arginine is actually sensed.  Nevertheless, breeders can already use this study by looking for variations of BAC2 and AAP5 that allow plants to senesce slower in response to stress.

Wanneer er geen weg meer terug is

Wanneer is het tijd om dood te gaan, dat is de achterliggende vraag van de laatste studie van een groep van Zweedse onderzoekers. Ze vonden dat het waarnemen van uitgeputte levels van het aminozuur arginine de cel vertelt om de handdoek in de ring te gooien.

Bladsenescentie en de daaropvolgende dood zijn een normaal onderdeel van een plantenleven. We zien het elke herfst in actie, bladeren die rood en geel kleuren voordat ze van de bomen vallen. Het gebeurt ook tijdens de normale ontwikkeling wanneer oudere bladen doodgaan en gedurende periodes van stress, zoals droogte stress.

Tijdens bladsenescentie degraderen cellen hun eiwitten, nucleïnezuren, lipiden, en celstructuren om de daarin opgeslagen voedingstoffen te recyclen. Deze transporteren ze vervolgens de cel uit voor gebruik door jongere organen of voor opslag. Dit alles hoeft niet automatisch in de dood van de cel te eindigen. Als bijvoorbeeld de droogte stress eindigt voordat de senescenesterende cel het punt van geen weg terug heeft bereikt, dan kan de cel zich herstellen.

Senescentie verhinderen

Dit roept de vraag op: hoe weet de cel wanneer het tijd is om te sterven? Planten veredelaars zien het antwoord op deze vraag als een manier om meer controle over bladsenescentie te krijgen. Dit, zo hopen ze kan helpen om opbrengstverliezen te verkleinen en het veredelen van gewassen mogelijk te maken die minder gevoelig zijn voor door stress geïnduceerde senescentie.

De eerste hint op het antwoord op deze vraag kregen de onderzoekers van een mutant die geen stress geïnduceerde senescentie heeft. De bladeren van deze plant accumuleren aminozuren in grote hoeveelheden. Om uit te vinden of een van deze aminozuren senescentie beïnvloed stelde de onderzoekers bladschijfjes bloot aan een bepaalde aminozuur. Hierbij vonden ze dat alleen in het geval van extra arginine het bladschijfje z’n chlorofyl hoeveelheid langer op pijl hielt vergeleken met de bladschijfjes die met andere aminozuren behandeld waren of de controle. Dit suggereert dat een afname van arginine onder de drempelwaarde signaleert dat het tijd is voor senescentie.

Herstellen van senescentie mogelijkheid

Vervolgens keken de onderzoekers naar de genen die arginine uit het cytosol transporteren. Voor twee van deze genen, AAP5 en BAC2, vonden de onderzoekers dat deze actief waren tijdens senescentie. Van deze wordt BAC2 zelfs direct geactiveerd door de senescentie master regulator. Vervolgens activeerde de onderzoekers deze arginine transporteurs in planten wiens bladeren geen bladsenescentie ondergaan. Dit herstelde het vermogen van senescentie.

Een belangrijke vraag nu is werkt dit ook andersom. Dat is, wanneer arginine levels hoog blijven vertraagt dit dan de senescentie tijdens de normale verouderingsprocessen. Dus de onderzoekers creëerde planten zonder AAP5 of BAC2 of allebei. Terwijl planten die een van deze arginine transporteurs miste geen vertraging van normale veroudering hadden, hadden planten die allebei deze transporteurs hadden dat wel.

Dus de hoeveelheid arginine functioneert waarschijnlijk als een bron die de cel vertelt bij welk stadium van senescentie het is. Maar zoals de onderzoekers ook zeggen er zijn nog meer details nodig. Zoals of de distributie van arginine in het cytosol dit alles beïnvloed. Of zoals hoe arginine eigenlijk wordt waargenomen. Niet te min kunnen veredelaars deze studie ook nu al gebruiken om te kijken of variaties van BAC2 en AAP5 het mogelijk maken om planten in hun reactie op stress langzamer te laten senesceren.

Plants, like all organisms don’t like an excess of salt. One of its ways to deal with salt stress is to strengthen their cell walls. FER, a receptor in the cell membrane is one of the key proteins in regulating this. Now a group of Chinese researchers show that key to this is the small molecule phosphatidic acid.

An excess of salts doesn’t only cause havoc inside cells but also cause damage to the cell wall. One of the ways it does this is through the binding of sodium ions to the cell wall. This loosens up the fibres and peptides which made up the cell wall. And one of these peptides, RALF, when it no longer is hold back by the cell wall binds to the FER which subsequently sounds the alarm.

After sounding the salt alarm, the plant gets to work on its cell walls. Not only to repair the damage but also to strengthen it. This is needed as cells placed in a high salt environment lose water as osmosis draws it out of the cell. With out strengthening the cell wall, the cell would collapse. In addition, strengthening the cell wall helps with preventing salt ions from entering the cell.

New interaction partner

Now a fair bit is known about how FER sounds the alarm. But the researchers had a feeling that more was going on. So, they did an assay to find out if FER binds to any other proteins than known so far. Finding that the membrane protein PLDδ interacts with FER. PLDs are proteins that produce phosphatidic acid. And as it turned out PLDδ was not the only PLD FER bound to, it also bound to PLDα1

Plants without both PLDα1 and PLDδ can’t handle to much salt, similar to plants without FER. And when plants miss all three of these proteins their ability to deal with salt stress is even further diminished.

The question now is why do FER and PLD interact. This the researchers found was not because the PLDs needed FER, as they first expected, but rather the other way around. FER needs PLD, or to be more precise FER needs the phosphatidic acid that is produced by PLD. When the researchers blocked PLD from producing any phosphatidic acid FER was quickly removed from the plasma membrane.

Finding the details

And this the researchers found out put the salt response, and subsequent cell wall strengthening in jeopardy. FER is actually needed to stay a bit longer at its place in the plasma membrane. However, that was not the only job of phosphatidic acid. Phosphatidic acid also promoted FER to do its job. One of part of which is to ensure that the proteins needed for strengthening the cell wall reappear at the plasma membrane after the initial salt stress shock.

Knowing that, in order to respond to salt stress FER needs those two PLD proteins, helps plant breeders keeping an eye out for them when looking for salt tolerant plants.

Continuing studying how plants deal with stress after the general mechanisms are known helps to identify all players involved. Only then can plant breeders with precision develop robust plants that respond predictable to stress.

Planten zoals alle organismen houden niet van te veel zout. Een van de manieren van omgaan met zout stress is het verstevigen van hun celwanden. FER een receptor in het celmembraan, is een van de belangrijke eiwitten die dit reguleren. Nu laten een groep Chinese onderzoekers zien dat een klein molecuul fosfatidezuur dit allemaal mogelijk maakt.

Een overdaad aan zout gooit niet alleen binnenin de cel alles overhoop, het zorgt ook voor schade aan de celwand. Zout doet dit doordat natrium ionen aan de celwand binden. Dit zorgt ervoor dat de vezels en peptiden van de celwand losser komen te zitten. Een van die peptiden, RALF, bind vervolgens aan de FER receptor die daarop alarm slaat.

Na geattendeerd te zijn gaat de plant aan het werk aan z’n celwanden. Niet alleen om de schade te repareren maar ook om ze te verstevigen. Dit is nodig omdat cellen in een zoutrijke omgeving water verliezen omdat dit door osmose de cel wordt uitgetrokken. Zonder de celwand te verstevigen zou de cel ineenstorten. Daarnaast helpt het verstevigen van de celwand ook met het buitenhouden van zout zelf.

Nieuwe interactie partner

Nu is er redelijk wat bekend over hoe FER alarm slaat. Maar de onderzoekers hadden het gevoel dat er nog meet te ontdekken was. Daarom deden ze een assay om uit te vinden aan welke andere eiwitten FER bindt. Dit bleek het membraaneiwit PLDδ te zijn. PLDs zijn de eiwitten die fosfatidezuur produceren. En het bleek dat PLDδ niet het enige PLD eiwit was dat aan FER bind, PLDα1 bindt ook aan FER.

Planten zonder PLDα1 en PLDδ kunnen niet veel zout aan, net zoals planten zonder FER. Maar dat valt in het niet bij planten die alle drie deze eiwitten missen, daar is de mogelijkheid om met zout stress om te gaan nog minder.

De vraag was waarom binden FER en PLD aan elkaar. Dit bleek zoals de onderzoekers uitvonden was niet omdat de PLDs FER nodig hadden zoals de onderzoekers eerst dachten, maar eerder andersom. FER heeft PLD nodig, of om op nauwkeuriger te zijn FER heeft de fosfatidezuur die PLDs produceren nodig. Wanneer de onderzoekers PLDs verhinderde om fosfatidezuur te produceren resulteerde dit in dar FER snel van het celmembraan verdween.

Details vinden

En dit, ondervonden de onderzoekers, brengt de reactie op zout en de daaropvolgende versterking van de celwand in het geding. FER heeft het nodig dat het wat langer op z’n plaats in het celmembraan blijft. Al is dat niet de enige taak van fosfatidezuur. Fosfatidezuur is ook nodig om FER te stimuleren om z’n werk te doen. Onderdeel daarvan is het zorgen dat de eiwitten die nodig zijn voor het verstevigen van de celwand terugkeren naar het celmembraan na de initiële zout stress schok.

Dit alles wetende, dat om op zout stress te kunnen reageren FER deze twee PLD eiwitten nodig heeft helpt veredelaars om een op ze te letten wanneer ze op zoek zijn naar zout tolerante planten.

Dit doorgaan met het in kaart brengen met hoe planten omgaan met stress al is het algemene mechanisme bekend helpt om alle spelers bovenwater te krijgen. Alleen dan kunnen veredelaars nauwkeurig en met precisie robuuste planten ontwikkelen die voorspelbaar op stress reageren.
